Am I right in assuming that the base method of conflict resolution is something like [d10 (+/-) Offensive Ability Score + Relevant Proficiency] vs. [d10 (+/-) Defensive Ability Score + Relevant Proficiency]? Or is multiplication involved somewhere?

Almost correct, so that's on me for lack of clarity. I'll need to make this more clear in the post.
It's actually;
[d10 x (Offensive Ability + relevant Proficiency)] vs. [d10 x (Defensive Ability + relevant Proficiency)]
And when it's vs. a DC then it's [d10 x (Relevant Ability + Relevant Proficiency)] vs. [DC#]
I've got visual aids and tables for this bit as well, so I'll get those up at some point.

Might want to save the Status Effect section for closer to the Combat "chapter" as it were.

Yeah you're right - I'm introducing certain things in what feels like related sections but would be better in a 'game play' section.

I think I'm confused as to the usefulness of Proficiencies. Namely, if they enhance the Ability modifier, why wouldn't it be more useful to just add to the Ability score directly? Or is it that they add more in a narrower context (i.e., the equivalent of a two Ability score unit bump in one subsection vs. a one Ability score unit bump generally)?

Not quite sure what you're asking here but I'll try to explain how this works a bit more:
Proficiencies are added to your 'Ability' modifier only when preforming a related activity. If you are Climbing a cliff face for example, you get to add your Climbing Proficiency to your Ability multiplier (the Ability in this case being your Grapple score).
Hope that helps. I'll be doing some more editing. These sort of questions/input are helpful in making this 'guide' a more polished final product, so thanks.
